3.Observation items
1.Recoed the nerve trunk AP
(1)Observing the biphasic AP
a.Keep the equipment in sampling condition.Give the specimen an upper-threshold stimulus as above
b.Observe the wave form of biphasic AP of sciatic nerve
c.Change the stimulation intensity. Observe the relationship between stimulation intensity and the amplitude of AP. Find out the min stimulation intensity(max stimulation intensity) that can cause the maximum AP.
d.Reverse the sciatic nerve specimen, observe the change of biphasic AP
e.Switch the electrodes, observe the change of biphasic AP
(2).Observing the monophasic AP: clip the nerve between two record electrodes by tweezers in order to injure the nerve. We can observe the monophasic AP
2.Calculating the conduction velocity of AP
Place another sciatic nerve specimen in the shield-box, induce the biphasic AP as follows. Click the guage,limit the cursor at the pseudo-track strating point to Ap strating point in order to get the time from giving stimulus to Ap appears.Measure the distance between two electrodes,input the data to computor,calculate the v.
3.measure the refractory period
(1).Adjusting to the maximum stimulation intensity, induce the biphasic AP
(2)Adjusting time intervals of successive single stimulus in order that two independent wave-forms of biphasic AP appears
(3)Lower the time intervals gradually, make the latter one fusing with the former one.When the latter one becoming small suddenly,record the point T2.It illustrates that it has already gone through the relative refractory period of the previous AP. Keep lower the time intervals,when the latter one disappearing suddenly,record the point T1.It meas that it has entered the absolute refractory period of the previous AP. The absolute refractory period is from pseudo-track strating point to T1. The relative refractory period is from T1 to T2
4.The influence of Procaine on sciatic nerve AP
Stimulate the sciatic nerve trunk with 2% procaine cotton.After 10 min,remeasure the conduction velocity, refractory period. Observe the change of conduction velocity, refractory period with the former.
[Notice]
1. Do not hurt the nerve during the separation in order to avoid the influnce of results.
2. the sciatic nerve specimen should be as long as possible,8cm above is the best. Wash it with Ringer’s solution on times to keep its good excitability.
3. Keep the nerve trunk in good touch with electrodes.The distance between two electrodes should be as long as possible.
